随着社会的快速发展,创伤性骨关节炎患者越来越多,给患者及社会带来沉重的负担。随着研究的不断深入,具有广谱蛋白酶抑制作用的α-2巨球蛋白(α2M)在创伤性骨关节炎及相关疾病的保护作用不断被发现。本文就此作一综述,详细阐述了α2M的功能,结构;α2M在创伤性骨关节炎中的早期诊断、保护作用;在膝关节交叉韧带损伤修复中的促进作用;在半月板损伤中的保护作用;在韧带重建中骨-腱愈合的促进作用。最后,本文指出了目前面临的一些挑战,如,α2M的具体作用机制,生物安全性需要进一步深入研究。

The growing population of post-traumatic osteoarthritis(PTOA) patients have brought great burden to both the patients themselves and the society. However, with the discover of alpha-2-macroglobulin(α2M), a broad spectrum of protease inhibitors, the greater protective effect has been found on post-traumatic osteoarthritis and relataed diseases. This article reviewed the structure and fuction of α2M, the main effect on early diagnosis and treatment of PTOA, the promotion during the repair of anterior cruciate ligament injury, the protective effects on meniscus injury and the positive effects on tendon-bone healing during anadesma reconstruction. This article also pointed out some of the major challenges including specific mechanisms of action and biosecurity of α2M, which need some in-depth studies in the future.
